چکیده

In order to obtain information on the mechanism of ether bond formation in the synthesis of 0-alkyl lipids, a study was carried out to determine if there is a loss of hydrogen from carbon 3 of dihydroxyacetone phosphate (DHAP) (nonphosphorus-linked carbon) in the course of its incorporation into glyceryl ethers. Accordingly, a mixture of [1,3-3H2] DHAP and [I ,3-WJDHAP was prepared. In a microsomal enzyme system from Tetrahymena jyriformis which synthesizes 0-alkyl lipids, it was found that there was a loss of hydrogen from carbon 3 of glyceryl ethers synthesized from double-labeled DHAP. The loss was quantitatively consistent with the Iabilization of one of the hydrogens linked to carbon 3 of DHAP and was not due to isomerase activity.

Biosynthesis of an O-alkyl analogue of phosphatidic acid and O-alkylglycerols via O-alkyl ketone intermediates by microsomal enzymes of Ehrlich ascites tumor.

Microsomal enzymes from Ehrlich ascites tumors synthesized 0-alkyl glyceryl ethers from hexadecanol and dihydroxyacetone phosphate in the presence of ATP, CoA, and Mg++. Glyceryl ether synthesis occurred via the formation of 0-alkyldihydroxyacetone phosphate and O-alkyldihydroxyacetone. Although NADPH inhibited the initial reaction of dihydroxyacetone phosphate with fatty alcohol, it was requir...

Exclusive localization in peroxisomes of dihydroxyacetone phosphate acyltransferase and alkyl-dihydroxyacetone phosphate synthase in rat liver.

Dihydroxyacetone phosphate acyl transferase (DHAP-AT), alkyl dihydroxyacetone phosphate synthase (alkyl-DHAP-synthase), and glycerol-3-phosphate acyltransferase (GPAT) activities were investigated under optimal assay conditions using highly purified organelle preparations. The data presented clearly indicate that GPAT activity was mainly localized in mitochondria and microsomes, whereas DHAP-AT...

Hydrogen exchange in the formation of dihydroxyacetone phosphate from acyl dihydroxyacetone phosphate in O-alkyl lipid synthesis in Ehrlich ascites tumor cell microsomes.

We have previously presented evidence that acyl dihydroxyacetone phosphate is converted to O-alkyl dihydroxyacetone phosphate via an endiol intermediate which then accepts a fatty alcohol to form O-alkyl dihydroxyacetone phosphate. We have further proposed that, in the absence of fatty alcohol, the endiol derivative of acyl dihydroxyacetone phosphate reacts with water to form dihydroxyacetone p...

Induction of the peroxisomal glycerolipid-synthesizing enzymes during differentiation of 3T3-L1 adipocytes. Role in triacylglycerol synthesis.

The glycerophosphate backbone for triglyceride synthesis is commonly believed to be created through the conversion of dihydroxyacetone phosphate (DHAP) by glycerophosphate dehydrogenase (GPD) to sn-glycerol 3-phosphate (GP), which is then converted by glycerophosphate acyltransferase (GPAT) to 1-acyl-GP. Consistent with this, GPD and GPAT are highly induced during differentiation of mouse 3T3-L...

Biosynthesis of glycerolipids via acyldihydroxyacetone phosphate.

Acyldihydroxyacetone phosphate was first discovered as a lipid rapidly labelled by ’’Pi or by [p”P]ATP in guinea-pig liver mitochondria (Hajra & Agranoff, 1967,1968). This rapid labelling was due to the dephosphorylation and rephosphorylation of endogenous acyldihydroxyacetone phosphate in mitochondria (Hajra et al., 1968). Acyldihydroxyacetone phosphate was shown to be biosynthesized by the di...
